چکیده

Gene suppression via RNA interference (RNAi) provides an alternative strategy for insect pest management. The ingestion by insects of double-stranded RNAs targeting essential insect genes can trigger RNAi and lead to growth inhibition, developmental aberrations, reduced fecundity, and mortality. This RNAi response is particularly acute in certain coleopteran species, most notably the western corn rootworm, a devastating pest impacting corn production in the United States. The development of nextgeneration rootworm-protected corn hybrids includes an RNAi-based trait that provides a mode of action distinct from those of Bacillus thuringiensis insecticidal protein-based traits currently used for rootworm pest management. Unfortunately, many insect species including important lepidoptera and hemiptera pests appear largely recalcitrant in their response to environmental RNA, suggesting biological barriers that thus far limit the utility of RNAi for agricultural pest management. This review will highlight recent Advances in Insect Physiology, Volume 47 # 2014 Elsevier Ltd ISSN 0065-2806 All rights reserved. http://dx.doi.org/10.1016/B978-0-12-800197-4.00005-1 249 efforts to understand the barriers to RNA delivery in recalcitrant insect species, describe recent advances in the commercial development of insect-protected crops and biological insecticides utilizing RNAi, and discuss this strategy in the context of an integrated pest management approach.

RNAi Efficiency, Systemic Properties, and Novel Delivery Methods for Pest Insect Control: What We Know So Far

In recent years, the research on the potential of using RNA interference (RNAi) to suppress crop pests has made an outstanding growth. However, given the variability of RNAi efficiency that is observed in many insects, the development of novel approaches toward insect pest management using RNAi requires first to unravel factors behind the efficiency of dsRNA-mediated gene silencing. In this rev...

Application of RNA interference ( RNAi ) by feeding to pest management

Mini review Application of RNA interference (RNAi) by feeding to pest management M1 Yuki Maruyama RNA interference (RNAi) is a powerful tool to specifically suppress gene expression in both invertebrates and vertebrates, therefore, it has not only been used widely to study gene function in insects, also thought to be available for application to pest management. However, practical utilization w...

Parental RNA interference of genes involved in embryonic development of the western corn rootworm, Diabrotica virgifera virgifera LeConte.

RNA interference (RNAi) is being developed as a potential tool for insect pest management and one of the most likely target pest species for transgenic plants that express double stranded RNA (dsRNA) is the western corn rootworm. Thus far, most genes proposed as targets for RNAi in rootworm cause lethality in the larval stage. In this study, we describe RNAi-mediated knockdown of two developmen...

The Insect Ecdysone Receptor is a Good Potential Target for RNAi-based Pest Control

RNA interference (RNAi) has great potential for use in insect pest control. However, some significant challenges must be overcome before RNAi-based pest control can become a reality. One challenge is the proper selection of a good target gene for RNAi. Here, we report that the insect ecdysone receptor (EcR) is a good potential target for RNAi-based pest control in the brown planthopper Nilaparv...

Next-Generation Insect-Resistant Plants: RNAi-Mediated Crop Protection.

Plant-mediated RNA interference (RNAi) shows great potential in crop protection. It relies on plants stably expressing double-stranded RNAs (dsRNAs) that target essential genes in pest insects. Practical application of this strategy is challenging because producing sufficient amounts of stable dsRNA in plants has proven to be difficult to achieve with conventional transgenesis. In addition, man...
